You need both. GlovePIE is a "translator" for many different hardware input devices. This includes joysticks, gamepads, mice, keyboards, MIDI input devices, HMDs, Wiimotes, trackers. You can manipulate signals in all different ways. I posted a "MIDI to Mouse" script some time ago.
MidiYoke is a virtual MIDI driver. Like a MIDI cables inside your computer. It is used to connect any MIDI application outputs to any other applications inputs.
In our case we set GlovePIE´s MIDI output to MidiYoke1 and MixVibes´s MIDI input to MidiYoke1, too.
Dont forget to enable MidiYOke in MidiOX device settings to monitor everything.
